He can certainly activate it if he really wants to, but if he's still face-up on the field on resolution, then the effect will be negated.

well the idea is he is in your hand when his effect resolves. So he won't be negated.

No, Mole will go back to the hand if the effect resolves properly.

So, if mole said, " Send this card to your hand to send 1 of your opponent's monsters to their hand." then you could run skill drain and it would work?

If Grand Mole's return to hand was a Cost, (and yes, your wording there does look like one) then Skill Drain would not affect it.
But it's not a cost. It activates on the field, and goes to hand on resolution - this will be negated by Skill Drain.
